Calculations of binding affinities for MGMT/inhibitor complexes with computer simulation techniques

The suicidal DNA-repair protein O6 -methylguanine-DNA methyltransferase (MGMT) reverses alkylation at the O6 position of guanine, thereby conferring resistance to alkylation cancer chemotherapies. It must be inhibited to sensitize tumor cells for chemotherapy and is therefore an important anticancer drug target. Recently, a number of O6 -benzylguanine-derived inhibitors were studied experimentally. Here, the absolute and relative binding affinities of these compounds binding to MGMT shall be determined using computer simulation techniques. The calculations will yield details of the binding modes and structures of the MGMT/inhibitor complexes. This will provide deeper insight into the structure-function relationship of MGMT and thus serve as an important step towards the development of more potent anticancer drugs.
